연구분야

N-end rule 경로는 특정 N-말단 잔기나 그 변형 산물을 분해신호(degron)로 이용하는 세포내 단백질 분해시스템이다. N-end rule 경로에서는 특정 N-말단 잔기가 번역 후 변형(post-translational modification)을 거쳐 분해 신호(degron)로 변환이 될 수 있다. 활성화된 N-말단 분해 신호(N-degron)는 인식 요소(recognition component)에 의해 인식되며, 인식 요소는 기질을 유비퀴틴 단백질분해 경로나 autophagy-리소좀 경로로 전달하여 분해를 유도한다. N-말단 분해신호 전구체(pro-N-degron)는 아르기닌화 (arginylation) 등을 포함하는 효소 연속반응에 의해 활성화된다. 예를 들면, ATE1 R-transferase는 N-말단의 aspartate (Asp), glutamate (Glu), 혹은 cysteine (Cys)에 불안정화 아미노산인 arginine (Arg)을 중합시킴으로서 분해 신호를 생성시킨다. 본 연구실에서는 N-end rule 단백질 분해 체계의 작용 원리를 정확히 규명하고, 동물 모델을 이용하여 이러한 단백질 분해 체계의 조절 양상 및 기능, 병리학적 요인까지 포괄적으로 이해하기 하기 위한 연구를 수행한다.
